Will you need a separate amplifier for this length? Or is the booster contained in the cable?
Product 39011 does contain a active booster inline. It does not require any extra components only 100ma to power the active chip.

If I connect this to a wall plate, can I then add another 16ft USB to go from the wall plate to a computer?
You would need to use an active booster or a hub to allow the distance requested. 16.5 feet is the natural maximum distance that a USB cable can run.
